Endophytic fungi from Brachiaria grasses in Brazil and preliminary screening of Sclerotinia sclerotiorum antagonists Scientia Agricola

ABSTRACT: Fungal endophytes of Brachiaria, a nonhost of Sclerotinia sclerotiorum, may harbor species with antagonistic effects against this plant pathogen. The objective of this work was to investigate the diversity of endophytic fungi associated with different Brachiaria species and hybrids and evaluate their potential to inhibit the plant pathogen S. sclerotiorum. Stem samples from 39 Brachiaria spp. plants were collected in pasture fields and experimental areas of three states of Brazil resulting in 74 endophytes isolated. Twenty-eight species were identified by sequences of the Internal Transcribed Spacer (ITS) and 18S rDNA regions. The process of biological nitrogen fixation (BNF), performed by symbiotic nitrogen fixing bacteria with legume species, commonly known as α and β rhizobia, provides high sustainability for the ecosystems. Its management as a biotechnology is well succeeded for improving crop yields. A remarkable example of this success is the inoculation of Brazilian soybeans with Bradyrhizobium strains. Rhizobia produce a wide diversity of chemical structures of exopolysaccharides (EPS). Fungal endophytes of Panicum maximum and Pennisetum purpureum: isolation, identification, and determination of antifungal potential R. Bras. Zootec.

ABSTRACT This study aimed to isolate and identify endophytic fungi from the forage grass P. maximum and evaluate their ability to inhibit the growth of plant pathogenic fungi. One sample from P. purpureum grass was also included. Surface disinfected stem fragments were used for endophytic fungal isolation. One hundred and twenty-six endophytic fungi were isolated, of which 118 were from P. maximum and eight from P. purpureum. Morphological characteristics and internal transcribed spacer (ITS) and 18S (NS) sequence comparisons identified most isolated endophytic fungi as belonging to the phylum Ascomycota, with Sarocladium being the dominant genus. Morphological and molecular differentiation of the pectinase producing fungi Penicillium expansum and Penicillium griseoroseum BJM

Two species from the genus Penicillium, Penicillium expansum and P. griseoroseum (Brasilian isolates) were characterized morphologic and molecularlly. Morphological variability was detected among isolates in regard to colony morphology and to conidia coloration. The molecular characterization was based on the RAPD markers, telomeric fingerprinting and ITS sequencing. A total of 78 RAPD primers were used and 8 presented differences in band patterns with 54% of the amplified polymorphic fragments. The monomorphic fragments of 600 bp (P. expansum) and 594 bp (P. griseoroseum) were amplified.
